I don't see that anyone has replied. So I'll take a stab at this - I'm just a beginner that likes to tinker. But, anyway - if you are talking about modern nibs sold on the internet as a reference than I would say that they are referring to the size of the feed. A 6mm or 5mm feed equal respectively to a #6 and #5. If you are asking about vintage nibs than it is a whole other ball game with each maker having its own nib nomenclature.

Nib sizes are notoriously different!

 

Modern nib units made by schmidt, jowo, & Boch have small and large sizes and I think one of them might call them 6 and 5. But I wouldn't hope for absolute relativity.

The numbers are a general size from a manufacturer, but each manufacturer has their own standards for sizes. Company A's #5 could be slightly bigger than Company B's #6.
